Easicult TTC

Easicult TTC dip slides are used for monitoring microbial contamination in industrial environments. The slide is covered on both sides with triphenyltetrazolium chloride (TTC) agar which allows the growth of most common bacteria. Use to detect the elevation of total bacterial counts.

Easicult TTC Intended use

Easicult TTC slides are intended for monitoring of bacterial contamination in various industrial environments. The test can be performed on-site, or the slides can be used as convenient transport media for samples.

The slide is covered on both sides with TTC Agar which supports growth of most common bacteria. The main significance of the test is that elevation of total bacterial counts can be detected.

Warnings and precautions of Easicult TTC

  • Do not use the product beyond the expiry date marked on the kit.
  • Do not touch the unused growth media.
  • Do not use the slide if you notice
    • discoloration or dehydration of the growth medium
    • detachment of the growth media from the plastic slide
    • evidence of microbial growth.
  • Because any growth on the Easicult TTC slide may be pathogenic, do not touch the used slide

Easicult TTC Storage

Store Easicult TTC at room temperature (1825C / 6477F) protected from draught, temperature fluctuations and light sources. Avoid storage near heat-generating appliances. Do not allow to freeze. The expiry date (year-month-date) is marked on the box and on the cap of each slide.

To avoid contamination, the growth medium should not come into contact with any other material than the one to be tested. On the other hand, it is important that the growth medium makes full contact with the material to be tested.

Interpretation of results (Fig. 6)

Cautiously remove the slide from its tube after incubation and determine the bacterial count (number of colony forming units, CFU) by comparing the density of growth on the slide with the model chart. If the sample was diluted, the dilution factor must be taken into account in the evaluation. For example, if a dilution of 1 + 100 ml (1 ml of sample in 100 ml of water) shows a density of 106 CFU/ml, the actual result for the sample is 108 CFU/ml. As no universally applicable limits exist, limit values have to be determined by experience. For bacterial contamination in coolants, the following guide may be used:

Most aerobic bacteria grow on TTC Agar as red colonies. Moulds and yeasts may also grow slowly on this medium. Even though the bacterial growth is almost always in the form of red colonies, any colourless colonies should also be taken into account when the density is estimated. In cases where large colonies are present, it should be borne in mind that colony density, not the size of individual colonies, should be considered. If the bacterial count is very high (more than 107 CFU/ml), the growth is confluent. This can appear as a uniformly red surface layer. Very rarely there is totally colourless growth. It is advisable to compare slides exhibiting an apparently uniform surface with an unused slide to avoid misinterpretation
